To restore the immune cells’ clean-up function 🧹, we “switched off” glycolytic metabolism by removing hexokinase-2 bound to the mitochondria. This spurred microglia to gobble up 🍽️ toxic proteins that can build up in the brain 🧠 leading to Alzheimer’s. 🧵 (6/7)

We developed an optogenetic strategy to remove hexokinase-2 from the mitochondria in living cells to test this. Using this, we could spur microglia into clean-up mode with a flick of the light switch !💡 🧵 (5/7)

These damaged microglia relied on glycolysis 🍬 for energy⚡. The switch to glycolysis was controlled by binding of the glucose metabolizing enzyme, hexokinase-2, to the energy-producing 🏭 part of the cell, mitochondria. 🧵 (4/7)

In microglia, we found that the inflammatory marker, the translocator protein #TSPO, is critical for efficient energy production ⚡ and phagocytosis🍽️ Microglia lacking TSPO resembled damaged microglia observed in aging 👵 and Alzheimer's. 🧵 (3/7)

The immune cells of the brain, #microglia, need a lot of energy 💪 to clean up toxic waste ☠️ that can lead to #alzheimers 🧠 How is this fueled? 🔥 🧵 (2/7)
