AO-252 reported an 80% clinical benefit rate in the twice-daily dosing cohort, with 80% of evaluable patients achieving tumour stabilisation or regression lasting six months or more.
Importantly, these patients had already received a median of five prior lines of therapy.

Coiled Therapeutics has completed enrolment across two initial dose-escalation cohorts evaluating our new lipid-based formulation of AO-252 in our ongoing Phase I study.
🧬 Six patients enrolled: Three in each cohort, evaluating 120mg twice daily and 160mg once daily, with all progressing through their assessment period as planned.
🧬 Optimising drug exposure: The new softgel formulation is designed to improve absorption consistency and deliver more predictable drug exposure.
🧬 Advancing clinical development: We continue to target dosing up to 30 patients and expect initial safety and pharmacokinetic data by year-end 2026, ahead of planned Phase I/II expansion cohorts in ovarian and prostate cancer.
"This is an important step in de-risking our dose-escalation strategy and keeps us firmly on track to dose up to 30 patients by the end of the year, as we advance towards our planned expansion cohorts in ovarian and prostate cancer," said CEO Sridhar Vempati.
