delighted to have worked with @lab_peters @teichlab @Muzz_Haniffa @ClatworthyLab on this study. great work by first authors Emily Stephenson, Lisa Dratva and Erin Macdonald-Dunlop. Thanks to all our patients most of all
Our study on severe #COVID19 in end stage kidney disease patients with matched pre-pandemic samples from a broad cohort of 61 individuals is on medRxiv! medrxiv.org/content/10.1101/…
Evaluation of clonal T cell dynamics showed that the fastest expanding clones were significantly enriched in known SARS-CoV-2 specific sequences and shared across multiple patients. Our analyses also revealed novel TCR clones that are likely reactive to SARS-CoV-2.
We identified gene expression signatures of severity, with the majority of pathways differentiating mild from severe disease in B cells and monocytes.
We used single-cell RNA-seq, CITE-seq and immunoreceptor sequencing, derived from 187 longitudinal samples from 61 patients.
We collected samples before and during COVID-19 infection from the same individuals. We performed dense longitudinal sampling during infection, allowing us to study intra-individual changes over time among patients with a wide spectrum of COVID-19 severity.
We studied two cohorts of end-stage kidney disease patients prior to the introduction of vaccination. This is one of the patient groups most at risk from severe COVID-19.

In this study, interferon-γ was found to play a large role in the pathogenesis of APS-1. Results were confirmed in studies in animals and led to a trial of ruxolitinib in five patients, who had dramatic responses. Read the full study results: nej.md/4bXoKkG

ALT Original Article The Role of Interferon-γ in Autoimmune Polyendocrine Syndrome Type 1 Panel A shows photographs of hair on the scalp of Patient 1 before and after ruxolitinib treatment. The graph shows serum levels of interferon-γ before and after treatment. Panel B shows the macroscopic appearance of gastric mucosa during endoscopy in Patient 2. The graph shows serum levels of interferon-γ before and after treatment.

it's on the role of EROS in endothelial cells - our favourite protein takes on some different roles outside the immune system!
Delighted to share our new paper - a fantastic collaboration with Markus Waldeck Weiermair, Thomas Michel and the brilliant Michel lab at Harvard
