How are mitotic chromosomes built? 🏗️ Our Review @TrendsGenetics revisits the classical chromosome scaffold model and connects it to condensins, topoIIα, and physical forces. We propose interphase chromatin domains as “building blocks”🧩of chromosomes:

Great work Kazu! So happy to see it published so high!
Is euchromatin really “open”? 🧬 Using super-resolution imaging🔬 our new study @NatureGenet reveals: Euchromatin forms condensed domains in live cells. Cohesin constrains them and prevents domain mixing for proper transcriptional insulation🚧 🔗nature.com/articles/s41588-0… (1/2)
Excellent work! Particularly intrigued by the observations on nucleolus-associated heterochromatin.
Our new preprint @biorxivpreprint, led by @katsu_s_minami et al.! 🎉🧬🔬

Machine learning-assisted Repli-Histo labeling and single-nucleosome imaging reveal distinct transcription-dependent chromatin constraints across euchromatin and heterochromatin.
I am not afraid to say it: this is the most fundamental advance in our understanding of chromatin structure since the zig-zag fiber model.
Replying to @kazu_maeshima
Using single-H1 imaging and computational modeling, we show that most H1 behaves like a liquid inside chromatin domains, rather than binding stably to nucleosomes as in the textbook model.
Congrats, @MasaAShimazoe & all contributors! 🎉 2/2

🧬My retrospective chromatin review is out@Proc Jpn Acad: doi.org/10.2183/pjab.101.020
Textbook models of 30-nm fibers are fading. A new paradigm: chromatin forms liquid-like domains—locally dynamic, yet globally stable at chromosome level (viscoelastic)—supporting genome function.✨

Single-nucleosome imaging reveals that condensins act as molecular crosslinkers, locally constraining nucleosomes to organize mitotic chromosomes. Nucleosome-nucleosome interactions constrain whole chromosomes.
