So @adamfeuerstein -there was no benefit in overall survival with the addition of the mRNA vaccine and “… the question about what exactly was driving the vaccine’s benefit — was it the specific targeting of the neoantigens, or was it the general immune boost that studies have suggested mRNA vaccines generally provide?” This suggests that the immune reaction to mRNA vaccines is actually driven by the response to the LNPs and not the mRNA target…. what follows is the obvious question: What is the future of this platform and technology ? Maybe this might explain investors’ reaction…
About the intismeran presentation at #ESMO26 $MRNA $MRK My impression of discussant talk given by Inge Marie Svane of the University of Copenhagen was that she was positive if not overly enthusiastic about INTerpath-001. "Measured" might be the best, single adjective. "Brutal" is how an investor in the audience described the tone of her talk to me. (He was short Moderna, so there's that.) I disagree. Brutal is too harsh. If people were expecting Svane to jump for joy, then yes, she did not meet that expectation. To me, she played it mostly straight down the middle. She took no position on the mRNA vaccine vs neoadjuvant debate. (Slide below) She noted the lack of intismerna OS benefit, but said the same about neoadjuvant therapy. She also noted greater toxicity associated with neoadjuvant. For intismeran, Svane also raised the question about what exactly was driving the vaccine’s benefit — was it the specific targeting of the neoantigens, or was it the general immune boost that studies have suggested mRNA vaccines generally provide?

These curves don’t seem as impressive as the media campaign and company press releases reported back in August 🤔

Moderna shares dropped as much as 25% on Thursday following the leak of partial data on its closely watched cancer vaccine developed with Merck. Shares of both drugmakers fell even though the full data, released later Thursday following the leak, appeared meaningful for patients and positive relative to Wall Street expectations. cnb.cx/4i9eZoj

An important new analysis: 920 chemicals with breast cancer–relevant biological activity. 500 may be in consumer products, 462 in food, 162 in drinking water. No one can realistically avoid 920 chemicals. The opportunity? Prioritize what matters most and prevent exposure at the source. doi.org/10.1021/EHP.6c00022 @SilentSpringIns @EPA @NIHDirector_Jay @NIH @EHPonline

SARS-CoV-2 evolved since 2019 to evade human biological defenses. Obviously. But not just how it’s been known previously. Also to evade our *Hardwired* human biolog. That is - Not to just evade our prior immune responses - but actually to better evade eliciting a robust response by our naive B cells. Looking at prepandemic cryopreserved samples, @jbloom_lab showed a 3x difference in the fraction of naive B cells responsive to the original Wuhan spike RBD protein compared to more contemporary Omicron lineages. This isn’t a simple “evade prior elicited antibody response” as we all understand This is actually - the virus mutates to help avoid detection altogether by humans - uniquely by evading aspects of the human germline encoded naive B cells responsive repertoire. They look at mice, for example, and saw no difference in ability of naive B cells to respond to the different variants Altogether it points to a world where we not only have immune imprinting to impair protection, plus the “regular” virus mutation to avoid existing immunity, but now mutation specifically to avoid hardwired human biology.
In new study, we show that SARS-CoV-2 has evolved decreased inherent antigenicity in humans, as quantified by a ~3-fold decrease in the number of naïve human B cells that can bind the spike RBD. doi.org/10.64898/2026.10.15.…

This is a very interesting study from Fred Hutchinson Cancer Center. It suggests that SARS-CoV-2 may be evolving in a way that makes it increasingly difficult for the human immune system to recognize the virus in the first place—not simply harder for existing antibodies to neutralize it. So now one has to wonder whether if fewer naïve human B cells can recognize an evolved virus, will the vaccines even work? Will they have a difficult time recruiting genuinely new B-cell specificities? Doesn't this seem like an issue worth considering @FDADrugs @FDA @CDC @NIHDirector_Jay? “In a reanalysis of published data and in an independent cohort, after adjustment for multiple confounders, SARS-CoV-2 mRNA vaccination did not improve immunotherapy-related survival in patients with cancer.” “Together, these findings indicate that the previously reported vaccination survival advantage is largely explained by selection bias, with patients who had more favorable prognoses more likely to receive the SARS-CoV-2 vaccination, particularly in the early pandemic.” aacrjournals.org/cancerdisco… @weldeiry @RobertKennedyJr @VPrasadMDMPH @NIHDirector_Jay @jsm2334
