Have you ever wondered how chromatin states shape developmental trajectories? In our preprint, we address this question using single-cell nanoCUT&Tag, a method developed by Marek Bartosovic @marekbartosovic, biorxiv.org/content/10.64898…
We tested this idea by partially depleting H3K27me3 in the mesoderm, which led to reactivation of genes normally expressed in closely related cell populations.

ALT Epigenetic landscapes for the Krüppel gene locus in wild-type embryos and embryos with a mesoderm-specific knockdown of Polycomb catalytic subunit E(z), which deposits H3K27me3. After knockdown, the repressive "hill" diminishes along the muscle (M) trajectory, and Krüppel is ectopically expressed in muscles.

Using PRO-seq and CUT&Tag, we found that CBP activates zygotic genes in two steps:
- It helps recruit RNA Pol II to promoters without requiring catalytic activity.
- Its catalytic acetyltransferase function then triggers pause release.

Want to understand how gene expression programs form during development? Read our latest paper @GenomeBiology. We have profiled tissue-specific transcriptional and epigenomic landscapes forming as cells adopt distinct fates during Drosophila embryogenesis. rdcu.be/du0FU
To explore the dynamics of DV gene induction, we collaborated with the @sandberglab to infer gene burst kinetics transcriptome-wide from scRNA-seq data. We find DV genes are characterized by the capacity for large bursts of transcription, but with tightly controlled frequencies.
A big thanks to Rickard Sandberg and Christoph Ziegenhain @chris_zie for the nice collaboration on bursting and to Johan Reimegård @johan_rei for all bioinformatic help. It’s been a true pleasure to collaborate on this exciting project!
