Scientists at Mount Sinai have uncovered the first high-resolution structure of NBCn2, a brain protein linked to epilepsy, autism spectrum disorder, and other neurological conditions, and developed the first compounds capable of inhibiting its activity. Published in @NatureComms, the study used cryo-electron microscopy to capture the first highly detailed structural images of NBCn2, revealing how it transports sodium and carbonate ions that help regulate cellular acidity and control brain cell activity. The researchers also found that the way NBCn2 binds these ions differs from related proteins, suggesting that even closely related transporters may operate in distinct ways. The findings could help researchers better study disorders involving excessive neuronal activity and provide a new framework for understanding how altered brain signaling contributes to disease — potentially advancing future therapeutic research. Artificial intelligence and machine learning are being embedded in every aspect of the drug discovery and development process. Despite AI’s use at each step, the billion-dollar question is whether it can bring a larger number of novel drugs to market. cen.acs.org/physical-chemist…

A postdoc position is open in my lab to develop AI/ML models for predicting functional effects of genetic variants and to lead large-scale PheWAS in major biobanks. Strong programming skills, a solid publication record, and a background in human genetics are required.

Today, @NIH announced a new policy to cap how much publishers can charge NIH-supported scientists to make their work publicly accessible. This reflects our broader effort to restore public trust in public health by creating an open, honest, and transparent research atmosphere. Read more ➡️ bit.ly/4nCdIbl

Maybe @Google or @GoogleDeepMind should fund CASP, given their ample resources and how much they've benefited from the efforts of John Moult, Krzysztof Fidelis, Andriy Kryshtafovych, Nick Grishin, and many others. science.org/content/article/…

Unsolicited advice to new PIs and my opinion only not fact: As a new Assistant Professor, you have a 1-1.5 year window where you can get an R01 without a corresponding author paper IF you have a high impact 1st author paper from your postdoc. After that, it seems no amount of data in your R01 application matters anymore. The study section is looking for a peer reviewed publication. The important thing here is that it does not need to be high impact. They are looking for execution. Can you deliver the final product? “I don’t have time. Is there an alternative?” Put paper on preprint server and get it sent out at a respectable journal and note that in your R01 application. The study section may let this slide. Again, my OPINION based on my own experience. Good luck to all new PIs I know it’s a tough time.

Machine learning-based benchmarking of AI-powered docking for virtual screening Efficiently predicting how a small molecule will bind to a protein—a technique known as molecular docking—is critical for speeding up early stages of drug discovery. However, conventional physics-based docking can be slow and often struggles with large chemical libraries. Gu et al. compare these classical approaches with new AI-driven docking tools, investigating not just how accurately they reproduce crystal poses but also how well they enrich active compounds in virtual screening (VS). This multi-perspective benchmark helps identify potential strengths and flaws in both physics-based and AI-based methods, guiding researchers on where and how to deploy machine learning in large-scale compound discovery. The authors assembled “VSDS-vd,” a suite of datasets emphasizing real-world complexity: (1) The TrueDecoy set pairs experimental actives with genuinely assayed inactives, posing a difficult scenario for discriminating potent binders. (2) The RandomDecoy set intermixes actives with random compounds from a commercial library. (3) The MassiveDecoy set scales up to seven million decoys, mirroring industrial-scale VS. By running redocking trials, they compared CarsiDock, KarmaDock, DiffDock, and FlexPose (four AI docking tools) against four physics-based methods like Glide and LeDock, plus AI-based scoring (RTMScore, EquiScore). Pose quality was judged not just by RMSD but also by “physical plausibility” (using the PoseBusters toolkit), shining light on previously overlooked issues like protein–ligand steric clashes. Interestingly, top AI models (CarsiDock, KarmaDock) outperformed traditional methods in redocking success rate, yet physics-based tools usually offered more physically coherent poses—particularly in intermolecular checks. For actual VS tasks, physics-based Glide excelled on a challenging “TrueDecoy” scenario, whereas AI-based methods performed best on the more realistic “RandomDecoy,” suggesting diverse roles for different tools in practice. The authors found that postprocessing AI-generated poses (for example, with force fields) only modestly improved physical plausibility but did not necessarily help in ranking. Finally, they propose a hierarchical pipeline, using a swift AI approach (KarmaDock) for initial screening, then refining hits with a more accurate AI approach (CarsiDock) for final selection. This synergy can accelerate ultra-large VS campaigns while maintaining robust enrichment of promising drug leads. Paper: nature.com/articles/s42256-0…

98% of NIH grants that should have gone out this month, didn't. Despite court orders 'unfreezing' the funds, they are frozen. Biomedical research will grind to a halt. Clinical trials will end. All for no reason: the total impact on the US budget is miniscule

Misunderstanding on NIH funded research here. Grant proposals are reviewed by peers. Currently, only 10 out of every 100 proposals are funded, often after multiple resubmissions. Projects need to show strong progress to maintain funding. NIH funding has remained flat for decades.
Replying to @madamscientist
This is an estimate. What about all the negative impact projects of the governments choosing? What is the magnitude of those? It is incomplete to ignore the costs of taxes and then again the ignore the costs of gov’t malinvestment.
