🎗️ Selected this month by the Juisci HCP community: can first-line therapy move beyond trastuzumab in HER2-positive gastroesophageal adenocarcinoma? Zanidatamab, a dual HER2-targeted bispecific antibody, plus chemotherapy both with and without tislelizumab (anti-PD-1), showed encouraging efficacy as first-line therapy in phase 2 studies. HERIZON-GEA-01 (phase 3, New England Journal of Medicine) randomly assigned patients with previously untreated HER2-positive advanced gastroesophageal adenocarcinoma in a 1:1:1 ratio. Progression-free survival was longer with zanidatamab-tislelizumab-chemotherapy (12.4 months) and zanidatamab-chemotherapy (12.4 months) than with trastuzumab-chemotherapy (8.1 months; P<0.001), and overall survival was longer with the triplet (26.4 vs. 19.2 months; P = 0.004).
