I admit i am very much into intismeran…and it seems many others too as it is standing room only here…
Fabulous presentation/data on the Interpath-001 trial demonstrating clear benefit of this space age bespoke neoantigen vaccine along w pembro for pts w resected high risk melanoma as to 40% or so improvement in RFS/distant met free survival
Indeed a landmark trial for precision medicine and hopefully not too Long before we see confirmation in nsclc/other trials!
Interpath- indeed makes us foresee a wonderful path and light ahead for our field!

Landmark moment in personalized inmunoTx for cancer: individualized mRNA-based vaccine added to pembro prolonged RFS vs pembro alone in resected melanoma: standing ovation @myESMO #ESMO26 #ESMOAmbassadors @ProfGLongMIA gave a fantastic presentation! @EvanHallMD @FunchainMD
A long-awaited breakthrough in cancer vaccines.
One patient. One tumor. One personalized mRNA vaccine.
INTErpath-001 brings this vision closer to clinical reality. Are we ready to deliver it?

👑 Presidential #ESMO26! INTerpath-001: personalized mRNA vaccination reaches phase III in melanoma.
Resected stage IIB–IV melanoma, N=1,137: intismeran + pembrolizumab vs pembrolizumab.
💥 At 24 months:
• RFS: 82.2% vs 70.0% → HR 0.58
• DMFS: 88.5% vs 80.2% → HR 0.62
• Absolute gains: +12.2% RFS / +8.3% DMFS
• Benefit emerges mainly after 6 months
• Grade ≥3 AEs: 23.6% vs 20.3%
• OS still immature
A clinically meaningful new mechanism, but the real question now is positioning 🤔
For macroscopic stage III: adjuvant personalized vaccine after surgery, or neoadjuvant ipi/nivo before it? Biomarkers may ultimately decide.
Congrats to @ProfGLongMIA and all the team! 👏🏼

Important study by @ldiaz1971 @MSKCancerCenter advocating for universal germline testing after a cancer diagnosis. Among ~40,000 patients, 18.4% of early-onset, 15.6% of average-onset, & 12.3% of late-onset cancers harbor pathogenic germline alterations.
