I'm the Mediator Complex but you can call me MED. I'm essential for gene transcription & my best friend is the RNA pol II. And I have a head, a body & a tail.

Excited to share a preprint on the structural mechanism of transcriptional activation by human Mediator, studied using integrative structural modeling. Many thanks to Lacra Bintu @BintuLacra for helpful discussions! biorxiv.org/content/10.64898… @generegulation @MED_COMPLEX

I am a hot target
Thrilled to share my freshly "baked" Baker lab paper. We took a bottom-up approach to designing transcriptional regulators. Instead of relying on natural activation domains, we designed compact proteins that recruit specific cofactors. Activators for now, repressors to come!

We discussed how perturbing the Mediator Complex, a key gene expression regulator, reshaped cellular phenotypes. Some perturbations induced autonomous dedifferentiation, while others led to drug-resistant states. Interestingly, their effects aligned with the complex’s structure!

We are excited to report the discovery of TIGRs, a widely-occurring RNA-guided system found in bacteria and their viruses. TIGRs consist of a peculiar repeat region which is transcribed into RNA and processed into multiple guide RNAs to direct TIGR-associated proteins to their DNA targets. science.org/doi/10.1126/scie…
