We released a commentary on Ioannidis et al (2021) as a preprint. To date the settling of Polynesian islands, they inferred split times using IBD segments shared between modern genomes. We show that their estimator is biased to be ca. 300 years too old. doi.org/10.1101/2022.12.01.5…

Just how poorly does a polygenic score for LDL-C perform in a population that isn’t represented in its construction? Come check out my poster at #ASHG22 this afternoon (Wed) 3-4:45 to find out and chat!

Congratulations Dr. Carlson and team!!!@jenccarlson
If @jenccarlson and her team had only been looking in genetic databases with people of European ancestry, they would have missed a big cholesterol discovery, reported this week in @HGGAdvances. bit.ly/3F1hnf6

They say deep learning is just curve fitting. But how good is it in curve fitting exactly? Are DNNs as good as, or even better than, classical curve-fitting tools, e.g., splines and wavelets? We studied this carefully in arxiv.org/abs/2204.09664 and found many cool insights 🧵 1/

Very bad news: Accurate substitution modelling assumes all variables are in the same units.
But it is most common for studies to examine food in grams/servings while adjusting for total energy in calories.
This. Breaks. Everything.

Interesting paper reporting a stop gain variant in BTNL9 associated with reduced HDL and increased triglycerides. This variant is seen ~22% of Samoans (with many homozygotes) but extremely rare (<0.01%) or absent elsewhere in the world.

Why do complex traits differ in their genetic architecture?
Surprisingly, we find that two simple scaling laws explain why the number, effect sizes and frequencies of causal variants differ between traits.

The strongest possible sample size justification is based on an explicit statement of the smallest effect size that is considered interesting. You can't know what the 'true effect size' is - so design a study to detect effects that matter lakens.github.io/statistical…
