The aggressive cancer cells featured in this image are undergoing cell death because of a selective inhibitor: doi.org/10.1172/JCI202218
Here, David G. Kirsch @DKirschMDPhD, Pete Hendrickson @pghendrickson & team @UofT & @DukeHealth establish a next-generation mouse model and identify Minnelide as a potential treatment for CIC::DUX4 #sarcoma, a rare and deadly cancer with no treatment options.

Your donation dollars in action! 🙌A #StandUpToCancer-funded #research team unveiled an exciting #clinicaltrial breakthrough that reduces risk of #cancer relapse by 43% – for 2 of the most common types of soft tissue #sarcoma in adults, 2 years after treatment. Learn more: bit.ly/4bH9BEI.
Huge thanks to @ConquerCancerFd and @quadwfoundation for giving me this opportunity and for funding sarcoma research. Elated to join the growing network of QuadW researchers both near and far.
Congratulations to this year's class of Conquer Cancer Grant and Award recipients! We can't wait to celebrate with you at #ASCO24! Full list of recipients: brnw.ch/21wKdIf

Excited to share our work on chromatin landscape remodeling in cloned embryos! 🐄🐄We started off w/ simple question: how does the somatic epigenome reprogram to prepare for the onset of transcription (EGA)? tinyurl.com/yc4wv76p

(1/5) Our paper on pooled genetic screening of TERT-mutated #glioblastoma is out in @NaturePortfolio @oncogenejournal! We analyzed data from 500 #clinical and ~1300 #preclinical samples. Graphical abstract in the thread!

CIC-DUX4 fusion-gene driven sarcoma (CDS) is a devastating cancer that primarily affects kids and young adults. Because of its rarity, primary tissues are scarce and clinical trials are not feasible. Can we use mice instead? A thread…
Expression of the CIC-DUX4 fusion oncoprotein mimics human CIC-rearranged sarcoma in genetically engineered mouse models biorxiv.org/cgi/content/shor… #biorxiv_cancer
So what did we learn? Well, for starters, CIC-DUX4 is a very potent oncogene which seems to behave as a neomorphic transcriptional activator in line with prior findings.

Physician-scientists are a dying breed going from 4.7% in '80s to ~1.5% today. Multiple factors contribute including flat/declining #NIH funding. Translational research is critical for advancing clinical care.

In SLC for Brad Cairns’ 25th lab anniversary. Was truly a humbling experience to be a part of great science with great scientists. We’ve spanned the gamut from biochemistry of yeast chromatin remodelers, zebrafish epigenome reprogramming, ZGA, and germline stem cells.
Thanks @HenningWillers for an energizing finish to a great night @DukeRadOnc research retreat
So humbled and honored to receive this award. Thank you @ASTRO_org for recognizing our work and supporting my physician-scientist aspirations
Pete Hendrickson, MD, PhD, has been selected to receive the 2022 @ASTRO_org Residents/Fellows in #RadOnc Seed Grant for his project "Ewing-like Sarcoma: Targeting the CIC-DUX4 oncogene through epigenetic regulators." @pghendrickson is mentored by @DKirschMDPhD – congrats to both!

Very impressive work! What compensates Dux in mouse ZGA has puzzled me for ~4 years. I generated Obox1/2/5/7 QKO, but failed to see apparent phenotypes. It turns out that Obox4 is the promising one! A big mystery is largely resolved! biorxiv.org/content/10.1101/…
Thanks @DukeRadOnc and @DKirschMDPhD for the incredible support and confidence. The environment @DukeHealth is optimal for physician-scientist trainees.
Congrats to @pghendrickson (RORS, NCI R38 scholar) and PI @DKirschMDPhD for their recently awarded DOD grant to investigate models, mechanisms and tx for CIC-DUX4 sarcoma; an extension of Dr. Hendrickson's PhD work that defined the role for DUX4 in embryonic development #RadOnc
