Replying to @aedwards02
@aedwards02 (@KiraGenBio) presents on Multiple knockouts with prime/base editors to make allogeneic CAR-T cell therapies that can resist all of the most critical TME suppressive signals
The solid tumor microenvironment doesn’t suppress T cells through one pathway.
It runs a coordinated defense system.
Hypoxia → adenosine accumulation
→ PD-L1 ↑ and TGF-β ↑
These signals converge.
Block one, and the tumor pivots to the others.
The result: Stealth-TKO CAR-T
✓ Six simultaneous edits
✓ Resistant to adenosine, PD-1, and TGF-β
✓ Resistant to GvHD and allo-rejection
✓ Durable tumor eradication in humanized mice
This is what systems engineering looks like in cell therapy.
Solid tumors are complex, adaptive systems and simple therapeutics are ineffective
We have to meet this complexity with intelligent design.
This thesis closes one chapter.
The next one starts at @KiraGenBio.

This GD2 CAR-T neuroblastoma paper gives our field something rare: genuine hope.
40% complete remission
78% alive at 5 years
Kids still cancer-free more 5+ years after one infusion
This isn’t a promise for “someday.”
Ex vivo solid tumor CAR-T is arriving — and it’s here now.
A major win for solid tumors; GD2 #CART mediated a 40% complete remission rate in children with high-risk neuroblastoma @NatureMedicine !

Just launched our revamped @KiraGenBio website.
We’re building multiplex-edited CAR-T that act like noise-cancelling headphones for cell therapies—ignoring tumor suppression so our cells keep working.

Congress has officially recognized July 16, 2025, as the 7th annual Glioblastoma Awareness Day (#GBMDay)!
Thank you to @LindseyGrahamSC, @RepRWilliams , and all the bipartisan cosponsors in both chambers for championing this effort. Read more: braintumor.org/news/national…

When are we going to see progress for CAR T in solid tumors?", I'm often asked. More to do for sure, but progress is clear.

For the first time, allo CAR-T therapy has OUTPERFORMED auto CAR-T in the same patients! This offers hope for faster, scalable solutions—and we’re excited about how it validates our approach @KiraGenBio to tackling TME suppression. Let’s dive into it. 🧵

This is why we think the time is now for @KiraGenBio to work on CART for indications like GBM, where we can build upon the success of pioneers like @carlhjune, @DrORourke2, @christinebrown, @MarcelaMaus, @BryanDChoi and more!
#Glioblastoma (GBM) is a devastating disease. But hope may be on the horizon. #CARTcelltherapy trailblazer @carlhjune shares how the revolutionary therapy has showed promise in recent trials involving #GBM patients.

CART for AID strikes again. Stiff person syndrome. Someone get this study to @celinedion and her team stat!
This is another success for CAR T cells in autoimmunity - this time for stiff person syndrome
This syndrome is driven by autoantibodies, causing unpredictable bouts of stiffness and disability as a result
This patient had severe disease, resistant to conventional treatments, for ~10 years
She was treated with a single infusion of B cell-depleting CAR T cells, which reduced autoantibodies and rescued her ability to walk:
