Engineering T cells for immunotherapy vs cancer had already had marked success in certain types. But there's a next generation in the hopper, as reviewed today @Cancer_Cell, that ought to help take the field much further into solid tumors cell.com/cancer-cell/fulltex…

Thrilled to share our new paper in Science! 🎉 We uncover how dietary fat–driven ceramide synthesis activates YAP-dependent regenerative programs to promote colorectal cancer metastasis, independently of primary tumor growth.congrats @SwagataGoswam16 doi.org/10.1126/science.adw8…

Pulling out all the stops vs pancreatic cancer. The recent FDA-approved, celebrated drug daraxonrasib, increased median survival by 6.5 months @RevMedicines That's a positive step but clearly not enough. This new @Cancer_Cell paper lays out a path for more. cell.com/cancer-cell/fulltex…

CDK4/6 inhibitors arrest the cell cycle by activating Rb. But the authors provide compelling evidence that the story does not end there. A new Nature study shows that hypophosphorylated Rb unexpectedly enhances ER-mediated transcription. Rb relocates to enhancer regions and reactivates proliferative ER-target genes, including CCND1. So, paradoxically, CDK4/6 inhibition leads to: E2F ↓ → proliferation ↓ but at the same time Rb → ER transcription ↑ → proliferative counter-response ↑ Endocrine therapy shuts down this second pathway. This finding reveals a new mechanism that may help explain the strong biological rationale for combining CDK4/6 inhibition with endocrine therapy, while also pointing to a potential mechanism of resistance. nature.com/articles/s41586-0…

Our paper is out in @Cancer_Cell! We started this project almost 5 years ago with the idea that to understand why some cancer patients respond to immunotherapy and others do not, we may need to look beyond the tumor before treatment. Immunotherapy is not just a test of a pre-existing state; it is a perturbation. It changes the tumor immune microenvironment. So the question became: can the direction of that change explain response? authors.elsevier.com/a/1niWC…

#RareCancers face massive bottlenecks: limited patient material, few models, and slow screening pipelines. In our new Forum article in @TrendsCancer, we propose a "Zebrafish-First Triage Strategy" to rapidly model, test, and prioritize targets. cell.com/trends/cancer/fullt…

A holy grail for our lab has been tracking myeloid cells in human tumors in the same way that we track T and B cells with TCR/BCR. @vincentzliu and @CalebLareau solved it! We developed Mitotrek using scATAC-seq + mitochondrial DNA to do exactly this. Using Mitotrek, we find that new myeloid cells clones constantly infiltrate the tumor via circulating monocytes — and that their macrophage or dendritic cell fate is epigenetically programmed before tumor entry. @10xGenomics @parkerici @CancerResearch @TheMarkFdn cell.com/cancer-cell/fulltex…
