This
it's kinda cute that we're talking about "curing all diseases" when the best way to make an organoid is to just stick it inside an animal cells know the rules of how to make an organ, we largely still do not

It is our pleasure! As part of @TTIScience's Computational Biology program we're focused on building resources that cater to aging research, and protecting the investments the research community has made is part of that. – at Ottawa, Ontario
A few months ago, we faced some challenges keeping our online resources running due to a lack of funding. Thanks to the generous support of the longevity community, we were able to keep them online. We've now migrated our largest resources to a new server hosted by the Thalion Initiative, which will help reduce our operating costs and give us some breathing room while we seek additional funding. A huge thank you to @DToddWhite @MaxUnfried and @TTIScience for hosting our genome resources, including the bowhead whale and naked mole-rat genome resources, as well as GeneFriends, helping us continue to serve the research community. Thank you! 🙏

Scientists just estimated the upper limit of human lifespan determined by somatic mutations 👀 Researchers from Skoltech and AIRI developed a mathematical model to estimate how somatic DNA mutations influence the upper limit of human lifespan. Their model suggests that if nearly all other reversible aging processes were eliminated, the accumulation of DNA mutations alone could limit the median human lifespan to around 146-194 years, roughly twice today's human longevity. The model also found that somatic mutations reduced the median lifespan from a theoretical non-aging baseline of 1,759 years to around 156 years, highlighting how powerful mutation accumulation can be even in an idealized scenario. This study also identified brain neurons and heart muscle cells (cardiomyocytes) as the biggest longevity bottlenecks because they regenerate very slowly and accumulate mutations over time, while regenerative organs like the liver can replace damaged cells and maintain function much longer. Researchers conclude that somatic mutations are a major driver of aging, but they cannot fully explain human aging on their own, suggesting other hallmarks of aging contribute comparably to lifespan limits. These findings provide new insight into one of the fundamental hallmarks of aging and could help researchers develop future therapies aimed at extending healthy human lifespan.

Last week at Longevity Summit Dublin I spoke about Open Questions in Reproductive Aging and Female Longevity. The talk was inspired by @jpsenescence's paper mapping 100 open problems in aging against > 200,000 PubMed papers (post Thalion roadmapping workshop in Birmingham 2024).

Replying to @adamgries
@adamgries' last point is the key. Most philanthropy is unambitious, serves a tax purpose, and provides virtue signaling at the country club. Very few philanthropic entities are about advancing civilization and discovery or solving problems from first principles. 1/3
Replying to @brivael @elonmusk
Mostly, but not true for substantial parts of science. The Rockefeller Foundation helped launch modern molecular biology. The Wellcome Trust was a cornerstone of the Human Genome Project. HHMI funded scientists behind CRISPR and optogenetics. The Allen Institute created open brain atlases used by labs around the world. The existence of ineffective or poorly aligned charities doesn’t negate the extraordinary track record of philanthropic capital when it’s directed toward innovation rather than perpetual service delivery.

Sadly, in many cases the continuation of the philanthropic entity itself becomes the mission, as evidenced when foundations have dozens, hundreds, or even thousands of staff. In those environments it produces a bubble that insulates the principals from an understanding ... 2/3

Today I moderated this @theA4LI panel featuring several biotechs who are already using qualified state therapeutics access regimes such as Florida's Stem Cell Access law to bring treatment to patients, and others who are launching under Montana's Right to Try law shortly. We were also joined by @cole4nh and @KesselringSteve from the NH government, to discuss the future of their state's Right to Try programs. These state level pathways are critical to provide scalable patient access. Federal Right to Try and Expanded Access are important, but no single system is perfect, and inevitably patients fall through the cracks. Now, the states are there to step up and catch them when they do. If you're a biotech wanting to learn more about coming to Montana starting this July, my DMs are open!

My name is Stephen Martin. I am trying to get access to a nasal spray for this three year old boy, Brody DeVault. He has Creatine Transporter Deficiency. The creatine in his bloodstream does not make its way to his brain. Without treatment, he will live a life of intellectual disability, and probably die young from a seizure. There is a treatment which has finished Phase 1 clinical trials which may help him. I have spoken to the manufacturer, they will provide it to him under Montana's new law SB535. But only if we can get them official promises from HHS that they will not be punished for treating a patient under a state law. I have a meeting with the FDA on the 18th. Right now the single thing which most increases the likelihood of getting Brody treatment, is making sure that President Trump becomes aware of this before that meeting. I'm posting this in the hopes that I can bring this to the attention of people who can, in turn, bring this to the attention of President Trump. The meeting is in 6 days. Thank you for your attention to this matter!

Are biophysical constraints biology's underrated variable? We focus mostly on signalling molecules but this new paper from @PetrovaLab shows that a physical constraint like osmotic pressure can block lymphatic vessel growth even when growth factors are abundant. So it's not controlling it instead of signalling but alongside it like an independent gate. If we remove AQP1 (might have heard of this water channel, Nobel-winning discovery, etc) , gut lymphatics shorten, fat absorption fails, mice gain less weight on a high-fat diet. The physical microenvironment is a co-determinant in the process. Another curious thing is the context-specificity: AQP1 marks stress-adaptive lymphangiogenesis in the gut, lymphedema, and inflammation but not in normal embryonic development. We see different programs, depending on physical context. Maybe biology is more mechanical than the dominant frameworks suggest.

Later this month I will join a number of patients and biotechs who will be meeting with @US_FDA to discuss the importance of the department codifying its stance towards state Right to Try, Right to Personalized Treatment, and stem cell laws. Since 2016 when @SenRonJohnson first asked HHS and FDA to clarify their enforcement stance towards state programs, and was rebuffed, there has never been an official stance on this issue clarified. Yet, in the 10 years since, HHS and FDA have never actually escalated to enforcement against a doctor or biotech using a state Right to Try program. In one high profile example, they actually granted a drug priority breakthrough status after it was used successfully on 75 patients under Texas Right to Try. To some extent they have clearly been exercising enforcement discretion, which is the right thing for them to do! It's not a good use of their resources to go after companies sending medicine to patients for use under a state law. But, this position being "unwritten" is actively blocking patients from receiving care. And it's time for their position on the matter, whatever it may be, to be made official. These programs represent a chance to catch patients who fall through the cracks in the Federal patient access system. And the fact more and more states continue to pass new laws even today demonstrates a clear mandate from the American people to allow the states to step up to the plate, and get their patients medicine.
